NAA 98% Tissue Culture & Preparation Guide
Because NAA is only sparingly soluble in water, preparing a concentrated stock solution is recommended for accurate tissue culture dosing.
NAA should first be dissolved in a small amount of 95% ethanol or 1 N NaOH before being brought to final volume with distilled water.

Stock Solution Preparation

When to Add NAA to Tissue Culture Media
- Prepare your MS, basal, agar, or liquid tissue culture medium.
- Adjust the medium to approximately pH 5.6–5.8 before sterilization.
- Autoclave the culture medium normally.
- Allow the sterilized medium to cool to approximately 50 °C.
- Add the required amount of prepared NAA stock solution.
- Mix gently and thoroughly under sterile conditions.
Sterilize the culture medium by autoclaving first, then add NAA stock after the medium has cooled to approximately 50 °C when following protocols that treat the plant growth regulator as a heat-labile component.
